how to remove duplicates in source

please let me know how to remove duplicates while transfering source to target. i know that v have to use lookup stage but where to give condition .
suppose it is in this format

seqfile------------transformer------------target
\ \
\ \
\ \
lkp stage rejects

i mean where to do look up and how to use conditions
please let me know
