If a job is opened and left out then other users won't be able to access/open the same job. If i interpreted you question correctly.

Hi,
my question is job is opened and my connection lost, when I am try to open again its giving me the job is accessing by some other user .
In the above case if I logout from resources cleanup from directory itself
is there is any issues with respect to data stage repository or any affect to the same job

If your concern is about 'issues with respect to data stage repository or any affect to the same job' answer is No. Doing it from the director is the safest way (Esply if you are not aware of background processes).
